3. KEY TERMS & DEFINITIONS
To be "job-ready" in a cGMP environment, you must master the precise terminology used on the plant floor. Fluency in these terms is the first step toward professional competence and audit-preparedness.
- Combination Product Component: A part which is not associated with drug delivery that makes up the device constituent part of a combination product (e.g., finger flanges, safety devices, or backstops).
- Leak Integrity Qualified Material: Sterile, single-use processing assemblies (Sterility Level 3) that must be tested for leaks at predetermined, high-risk test sites.
- Primary Component: Containers or closures, such as syringes, vials, and stoppers, which directly hold the drug product and require visual and dimensional inspection.
- Raw Material: Chemicals or gases used in manufacturing that require chemical and/or microbiological testing and inspection.
- Sterile Qualified Material: Sterile, single-use product contact items that meet the requirements for Sterility Level 3.
- Supplier Quality Certificate (SQC): Any supplier-provided quality documentation (e.g., COA or Certificate of Conformance) associated with Combination Product Components and potentially primary components.

4. THE PROCEDURE, STEP-BY-STEP (WITH THE "WHY")

- Testing for Conformity: Every component must be tested against written specifications for purity, strength, and quality. Containers and closures must specifically meet the requirements of the item’s Material Specification Sheet (MSS).
- Why it matters: This prevents the use of substandard materials that could alter the efficacy of the drug product or introduce unforeseen impurities.
- The "3 Consecutive Lots" Rule: Each Combination Product Component type is qualified based on a specific Drawing number. If this number changes, qualification must restart with at least three consecutive supplier lots.
- Why it matters: A Drawing number change represents a change in the unique identifier of the component's specification; testing three lots ensures the supplier’s process is stable under the new design.
- Applying Specific Qualification Drivers:
- Elastomers (Stoppers/Caps): Qualification is driven by the specific stopper formulation provided in the SQC/COA.
- Glass (Syringes/Vials): Qualification is driven by the glass color (e.g., clear or amber) provided in the SQC/COA.
- Why it matters: Formulations and colors are critical to material compatibility and the protection of light-sensitive drug products.
- Prioritization of the SQC/COA: When multiple certificates are provided, the supplier’s SQC or COA must drive the qualification.
- Why it matters: The SQC/COA is the primary driver because it contains the manufacturer's validated results. Relying on secondary documents like packing slips is a compliance violation as they lack the "validation of test results" required by 21 CFR 211.84.
- Specialized Sterility and Integrity Claims: For sterile single-use items used post-filtration, reliability of the sterility claim must be qualified. For processing assemblies, leak integrity must be qualified at high-risk test sites.
- Why it matters: This ensures the highest level of sterility assurance for items that contact the drug product after it has been sterilized by filtration.
